So what is it? After 28 years of shooting I really think the 22 mag is my all time favorite. Good for rifles and revolvers plus a box of 50 fits in your pocket easily.
A "friend" has taken a lot of game with it in the past including deer. Cheap enough ($9 for 50) and its just about always on the shelves even during the ammo craze of '09. Shoots very well out to 150 yards and i'm working up to 200 for fun.

My favorite caliber to shoot is 9x19mm (9mm Luger). It's got enough punch to let you know you're shooting something, and can do some serious damage to targets. It's accurate enough out to 100 yards or so (for me anyway) and is relatively cheap ($9/50) so I'm not breaking the bank when shooting.

Id say 9mm, but the .22lr is a good one as well. I like the .22mag. I just wished there where more semi-auto offerings. I would love a semi-auto .22mag pistol and rifle.

Being a relative newbie to firearms, I'm only familiar with 9mm, 22lr, 223/5.56, 38sp, 357, and .410. I'd like to try out the 6.8SPC, 7.62x39, and the .50 Beowulf when and if funds allow, until then I'd have to agree with the others that 9mm offers the right combination of just enough punch yet relative affordability.

I'd have to say .45 ACP, as it was the first caliber I reloaded for, after discovering the greatness that is the 1911. 9mm is a close second these days, though.
