33 so far.

Edit: 38?

Edit again: 40, if there can be intersecting lines.

Hmm, some rules would help cause I know this is a trick.

I say there are 16 square spaces defined by lines with no other lines crossing through those spaces.

I count 40.

Don't get bent out of shape about the two that aren't in the grid. They only add 10 squares - The two big squares, plus the four on the inside of each one. Then you have 16 small 1X1 squares. Then you count 2X2 squares, starting on the top left and moving it one square to the right for each new square, then moving down one square and starting again, leaving you with 9 2X2 squares. Then you count 3X3 squares, which is only one for each corner since they're so large, giving you 4 extra. Then you have the one big 4X4 square.

10 + 16 + 9 + 4 + 1 = 40
